ISM brings together a community of organic and physical-chemical researchers interested in molecular structures, and working on their design, synthesis, characterization, reactivity and analysis in various environments. The Institute's expertise is based on a number of key skills: theorists in quantum chemistry, molecular and reaction dynamics, synthetic organic chemists, physical-chemical experimentalists (spectroscopy, photochemistry, molecular recognition and analysis), working in 10 research groups and 3 technical platforms.

As part of the Interdisciplinary and Exploratory Research (RIE) project GlycoMechano – Crosslinking of glycans to modulate the mechanical properties of the brain, we are recruiting a Post-Doctoral W/M whose work will focus on biological chemistry for the synthesis and imaging of non-natural sugars.

The aim of the GlycoMechano project is to develop molecular tools capable of modulating the rigidity of the neural extracellular matrix (ECM) of a neuronal tissue in order to study the effect of a rigidification of the synaptic environment on plasticity.

 Indeed, the molecular mechanisms of synaptic plasticity most studied are by far biochemical. However, the synapses are very motile and are surrounded by a dense and complex extracellular matrix. They are therefore subject to numerous mechanical constraints. The objective of the project will be to allow the visualization and manipulation of glycan chains of the extracellular matrix by metabolic labeling of unnatural oligosaccharides in neuronal tissues in order to study the effect of mechanical constraints on synaptic physiology.

Votre mission

Your mission will be to develop molecular tools and the cell labeling strategy from organic sugar synthesis to their visualization by fluorescence microscopy at various levels of resolution, including their validation in different cellular models. You will have to regularly present your research results (oral presentations to both teams, writing reports and publications) and carry out bibliographic monitoring on the subject.

•       You perform syntheses, purification and characterization of non-natural sugars for metabolic labeling

•       You carry out the synthesis, purification and characterization of bioorthogonal probes for imaging

# Cell culture and microscopy

•       You produce immortalized lines, dissociated neurons on mouse models and tissues, notably brain slices

•       You carry out fluorescence imaging shots: epifluorescence, confocal, dSTORM, DNA-PAINT, sptPALM ...

•       You analyze the images acquired with the software Fiji, Imaris, Matlab, ...

# Life of the laboratory and scientific dissemination

•       You prepare and host oral and/or written presentations of research findings at team meetings, institute meetings, activity reports, publications, conferences

•       You conduct a bibliographic monitoring

•       You contribute to the collective tasks of the laboratory: logistics, ...

Vos atouts / Vos talents

Holder of a PhD in Biological Chemistry, you have already worked on organic synthesis, preferably in glycochemistry. Knowledge in cell culture and fluorescence microscopy would be an advantage for this position.

More information:

In this position, you will work within the Molecular Photonics, Chemobiology and Imaging (PMCI) group whose research revolves around the development of molecular and analytical tools as well as dedicated technologies for chemobiology, molecular photonics, imaging, diagnosis and therapy. Our group is composed of two teams and a technical platform. You will be supervised by Frédéric FRISCOURT and work closely with Morgane Rosendale (IINS) - neurobiologist specialized in physiology and synaptic plasticity.

Based in Talence, Gironde FRANCE – access by tramway line B (stop « Peixotto ») buses, bike.

The laboratory is near the city-centre of Bordeaux and about 60 Km of the Atlantic coast.

12-month fixed-term contract

Gross monthly salary: 2750€

 

NB: the position is based in a laboratory in the Restrictive Zone, which requires an investigation prior to hiring, which can take up to 8 weeks.
